*Abstract:*
My lab is interested in elucidating the activity of virulence factors from
pathogenic bacteria so that we can gain novel molecular insight into
eukaryotic signaling. One of these factors encodes a Fic domain that
exhibits diverse activity depending on the species using this enzyme.
Studies with Fic domains reveal an unexpected complexity for
structure/function relationships that cannot currently be revealed using AI
but require empirical studies and insightful biochemistry. These studies
have led us to the characterization of Fic domains in eukaryotes, including
the metazoan FicD. In metazoans, the enzyme FicD is utilized for
AMPylation to maintain homeostasis in cells when under stress. Recently we
developed a pre-clinical model of dysregulated FicD AMPylation that causes
infant onset diabetes to elucidate molecular events altered in this
UPR-related disease.
